GHK-Cu Side Effects: What Topical Studies Report, and the Injected Unknowns

Only two small trials have tested GHK-Cu alone on skin, and copper is a weak allergen. Injected GHK-Cu has no human safety record, and the FDA flags its risks.

GHK-Cu is a small peptide of three amino acids bound to copper, and it is found in a long list of skin creams, serums and hair products. Its side-effect record is thin for a simple reason. Few controlled trials have tested it alone in people.

The record also splits cleanly by route. On skin, the concern is local irritation or allergy. By injection, the concern is everything that has never been measured. The difference between those routes is set out in GHK-Cu topical vs injected.

What topical trials recorded

A 2026 systematic review searched for studies testing GHK-Cu on its own for skin and aesthetic uses [1]. It found 20. Eighteen were preclinical, in cells or animals, and two were randomized trials in people.

One of those trials randomized patients who had laser resurfacing around the mouth to skin care with or without GHK-Cu [2]. Thirteen patients completed it. Blinded assessors found no difference in redness, wrinkles or skin quality, though patients using GHK-Cu reported higher satisfaction (P = .04).

Its published summary does not mention side effects. With 13 patients, the trial could only have caught a side effect that was common. A rare reaction would go unseen.

A separate 2025 review of GHK as an anti-wrinkle ingredient described “a surprising absence of clinical studies” of GHK-Cu, despite its wide use in cosmetics [3]. That absence is the main fact about its safety on skin.

Copper allergy

The copper in GHK-Cu raises the obvious question of allergy. A review of copper hypersensitivity pooled patch-test data from people with suspected symptoms or exposure [4].

A weighted average of 3.8% had a positive patch test to copper. The authors concluded that copper is a very weak sensitizer compared with metals such as nickel. They add that a few selected cases can have clinically relevant allergic reactions.

That figure is about copper in patch testing, not about GHK-Cu creams. No study has measured how often GHK-Cu itself causes contact allergy.

Injected GHK-Cu

Injected GHK-Cu is sold for recovery and healing, but no published study has recorded its effects in people. A 2026 review for orthopedic and sports medicine physicians found no clinical data supporting GHK-Cu for musculoskeletal conditions [5].

Immunogenicity means the immune system reacting to the product itself, or to impurities in it. It is a risk the FDA names; nobody has measured how often it happens with GHK-Cu.

What this means

On skin, GHK-Cu has a short, small record and a known, uncommon risk of copper allergy. By injection, it has no human record at all.
